摘要
研究了蚕丝固定化脂肪酶的工艺条件,并考察了固定化脂肪酶的稳定性。试验结果表明:蚕丝与对β硫酸酯乙砜基苯胺(SESA)进行反应的最适条件是pH=108,SESA:20g/g蚕丝,反应生成的对氨基苯磺酰乙基蚕丝(ABSE蚕丝)经重氮化后与脂肪酶偶联的最适条件是:pH=75,偶联时间>10h。加酶量为168~308u/g蚕丝时,所得固定化脂肪酶活力为106~160u/g蚕丝,此时固定化酶的活力回收率较高(>52%)。固定化脂肪酶稳定性较高,其操作半衰期约为250h。
The conditions of the lipase immobilized on the silk and the stability of the immobilized lipase have been studied.The results showed that the optimal conditions for the reaction of the silk with p β sulfuric acid ester ethyl sulfone aniline (SESA) are pH 10.8 and SESA 2.0g/g silk.The product p aminobenzent sulfanilyl ethyl silk (ABSE—silk) was diazotized and linked with lipase.The optimal pH for lipase linked to diao compound is 7.5.The link time is more than 10 hours.When the amount of lipase is between 168u/g silk to 308u/g silk,the activity of immobilized lipase is between 106u/g silk to 160u/g silk and the activity recovery is quite high (above 52%).The immobilized lipase is fairly stability,its operational half life is about 250 hours.
