摘要
目的观察周期性牵张应力对儿童骨髓间充质干细胞(MSCs)体外增殖活性与向成骨分化的影响。方法分离并培养儿童骨髓间充质干细胞,利用体外装置对3~6代细胞加载不同大小的牵张应力,相差显微镜观察细胞形态分布的变化,分别用细胞计数法、四甲基偶氮唑(MTT)和流式细胞仪检测细胞增殖、细胞周期及凋亡情况,同时检测碱性磷酸酶(ALP),骨钙素(OC)的活性。结果牵张应力干预MSCs后,细胞形态变长,沿受力环切线方向规则排列,细胞增殖能力增强,细胞周期比例改变,增殖指数增大,细胞凋亡则没有明显差别,ALP、OC的活性显著增强。结论适当的周期性牵张应力可以提高MSCs的增殖能力并促使细胞向成骨分化。
Objective To evaluate the effects of cyclical mechanical strain on the proliferation and osteogenic differentiation of pediatric bone marrow mesenchymal stem cells (MSCs) . Methods The pediatric MSCs were isolated and primary cultured. After the 3rd to 6th generation, MSCs were stimulated by mechanical stretch, the cellular morphology, distribution and alignment were observed. And cell counts, MTT assay and flow cytometry were applied to measure the cellular proliferation, cell cycle and apoptosis. The activity of ALP in cell and OC in culture fluid were measured. Results After mechanical stretch treatment, the cells had appeared slender and regularly aligned and evenly distributed porch stress circumcising line. At the same time, MSCs cellular proliferation had been accelerated, cell cycle had been changed, and the activity of ALP and OC had been increased. Conclusions These data suggest proper mechanical strain can enhance the cellular proliferation and osteogenic differentiation.
