摘要
目的研究白细胞介素-1β(IL-1β)、白细胞介素-6(IL-6)致痫与脑内信号分子protein kinase Acβ(PKAcβ)的关系。方法将实验大鼠随机分为4组:生理盐水组、IL-1β组、IL-6组、地塞米松组。行侧脑室注射相应试剂60min后观察大鼠行为学变化,并用SABC法显示各组大鼠大脑皮质及海马区PKAcβ的免疫反应变化。结果IL-1β组、IL-6组大鼠均有明显的痫性发作,且海马PKAcβ的免疫反应均明显强于对照组,差异具有显著性意义(P<0.05);地塞米松组大鼠呈抑制状态,且其脑内PKAcβ表达较对照组下降,差异有显著性意义(P<0.05)。结论在IL-1β、IL-6的致痫活动中,PKAcβ参与了其细胞内信号转导过程。
Objective To explore the effect of IL-1β or IL-6-induced seizure on the expression of PKAcβ in the brain of epileptic rats. Methods Rats were randomly divided into 4 groups: saline group, IL-1β group, IL-6 group and dexamethason group. The behaviors of the animals were observed 60 min after injection of corresponding reagents. The immunoreaction of PKAcβ in cerebral cortex and hippocampus of rats was measured by immunohistochemistry SABC staining method. Results Epilepsy was markedly attacked and immunoreaction of PKAcβ was obviously increased in IL-1β group and IL-6 group as compared with control group (P〈0.05). Epilepsy was markedly inhibited and immunoreaction of PKAcβ was obviously decreased in dexamethason group as compared with control group (P〈0.05). Conclusion PKAcβ may participate in intra-cellular signal transduction of epilepsy induced by IL-1β and IL-6.
