摘要
目的探讨全氟化碳乳剂(PFCE)静脉注射对急性肺损伤大鼠血清和肺泡灌洗液白介素-1β(IL-1β)水平的影响。方法56只雄性健康Wistar大鼠腹腔麻醉后分为对照组(NC组)、内毒素组(Lipopolysaccharide,LPS组)、PFCE预防组(预防组)。NC组静脉注射生理盐水10 ml/kg后2 h处死,LPS组和预防组分别静脉注射生理盐水/PFCE 10 ml/kg后10 min静脉注射LPS 8 mg/kg,于2 h4、h、6 h分批处死。比较三组肺脏病理损伤程度、血清和肺泡灌洗液IL-1β浓度。结果LPS组肺病理主要表现为肺泡隔明显增宽,大量白细胞渗出、聚集,部分肺泡腔中可见渗出液、出血、萎陷不张,预防组较LPS组明显减轻,病理损伤评分显著下降。与LPS组相比,预防组4 h6、h肺泡灌洗液IL-1β浓度[(0.08±0.01)ng/ml、(0.07±0.02)ng/ml]显著降低(P<0.05),而血清IL-1β浓度无明显变化。结论PFCE通过减轻肺脏炎性细胞浸润、降低肺内IL-1β释放可能是其预防急性肺损伤发生的重要机制之一。
Objective To assess the effects of intravenous perfluorocarbon emulsion (PECE) on IL- 1β expression in rats with acute lung injury.Methods 53 Wistar rats were randomly divided into normal control (NC) group, LPS (Lipopolysaccharide) group and PFCE group. NC rats were killed at 2 h after receiving intravenous 10 ml/kg normal saline, LPS group and PFCE group were treated with normal Saline or PFCE intravenously; after 10 min, all the rats were injured by intravenous injection of 7 ml/kg LPS. The extent of lung injury was assessed by histological examination, IL- 1β level in serum and broncho- alveolar fluid (BALF) at 2 h, 4 h, 6 h after injury. Results The degree of lung injury in PFCE group was milder than that in LPS group. Compared with LPS group, IL - 1β level in BALF of PFCE group decreased apparently at 4h and 6 h after injury. Conclusions Intravenous injection of PFCE attenuates lung injury and IL- 1β level in BALF of rats with acnte lung injury.
