摘要
目的研究血管内皮生长因子(VEGF)和增殖细胞核抗原(PCNA)在星形细胞瘤中的表达,探讨VEGF与肿瘤血管生成和细胞增殖的关系。方法SP免疫组化法对64例星形细胞瘤的VEGF和PCNA表达进行观察。CD31单克隆抗体免疫组化染色显示微血管,用微血管计数(MVC)测定肿瘤血管生成。结果VEGF蛋白主要分布在高级别胶质瘤组织的瘤细胞和内皮细胞,低级别胶质瘤呈低表达,且与PCNA的表达成显著正相关(r=0.991,P<0.01);高级别胶质瘤MVC显著高于低级别胶质瘤。结论VEGF可能参与胶质瘤血管生成,对胶质瘤的恶性进展起促进作用,PCNA较能客观地反映肿瘤的恶性程度。
Objective To investigate the expressions of vascular endothelial growth factor (VEGF) and proliferating cell nuclear antigen (PCNA) in human brain astrocytomas and its relation with angiogenesis and cell proliferation.Methods The expressions of VEGF and PCNA in 64 astrocytomas were examined by S-P immunohistochemical analysis, the vascular development was measured by microvessel counting (MVC) by immunosraining with CD31 monoclonal antibody. Results Immunohistochemical staining for VEGF showed the distribution of VEGF protein was mainly in tumor cells and endothelial cells of high-grade glioma, whereas it was only weakly expressed in low-grade glioma, and VEGF protein expression was significantly positive correlated with that of PCNA (r = 0.991, P 〈 0.01 ). MVC in high-grade glioma was considerably higher than those in lowgrade glioma( P 〈 0.05). Conclusion VEGF may contribute to the progression of human glioma and play an important role in leading to angiogenesis. PCNA was involved in malignancy of astrocytoma.
