摘要
[目的]研究尾悬吊模拟失重大鼠肺组织细胞凋亡情况的变化及其防护方法。[方法]采用尾悬吊模拟失重,分为悬吊7d、21d组和相应对照组以及悬吊7d地塞米松干预组,每组10只健康雄性SD大鼠,共50只。采用原位细胞凋亡检测技术检测肺组织的细胞凋亡情况,采用原位杂交技术检测肺组织BCL-2和BAX的表达情况。[结果]大鼠肺组织普遍存在凋亡细胞,7d尾悬吊模拟失重大鼠肺组织细胞凋亡水平明显高于对照组,差异具有统计学意义(P<0.05);21d组的差异无显著性(P>0.05)。7d尾悬吊模拟失重大鼠肺组织BCL-2 mRNA水平明显低于对照组,BAX mRNA水平明显高于对照组,差异均具有统计学意义(P<0.05);21d组的差异均无显著性(P>0.05)。地塞米松干预后细胞凋亡水平明显低于尾悬吊组,差异有统计学意义(P<0.05)。[结论]尾悬吊7d模拟失重大鼠肺组织存在细胞凋亡过度,凋亡调控基因也有相应变化;地塞米松可用于抑制模拟失重时肺组织的细胞过度凋亡。
[ Objective ] To investigate the changes and its protection of cell apoptosis in pulmonary tissues of tail-suspended rats in simulated weightlessness condition. [ Methods ] Experiments were conducted on 50 Spragno-Dawley male rats. The rats were divided into 5 groups: i.e. tail-suspended for 7 days group with its control group, tail-suspended for 21 days with its control group, and dexamethasone treatment at the 3rd day of tail-suspension group. Tail-suspension ( -30° ) was used as weightlessness simulation. The condition of cell apoptosis in pulmonary tissues was detected with in situ death detection, and the condition of BCL-2 and BAX mRNA in pulmonary tissues was detected by hybridization in situ. [ Results ] There were apoptotic cells in pulmonary tissues of rats in general. The level of cell apoptosis in pulmonary tissues of 7 d tail-suspended rats increased significantly ( P 〈 0.05 ), but that of 21 d group did not show a prominent change( P 〉 0.05 ). The level of BCL-2 mRNA and of BAX mRNA in pulmonary tissues of 7 d tail-suspended rats decreased and increased respectively with statistical significance( P 〈 0.05 ), but both those indices of 21 d group did not show a prominent change ( P 〉 0.05 ). The level of cell apoptosis in pulmonary tissues after dexamethasone treatment decreased significantly than that in tail-suspension group ( P 〈 0.05 ). [ Conclusion ] There were excessive apoptotic cells in pulmonary tissues of 7 d tail-suspended rats in simulated weightlessness condition and BCL-2 and BAX mRNA showed corresponding changes. Dexamethasone can be used to inhibit pulmonary tissues apoptosis in simulated weightlessness condition.
