摘要
目的:研究扶正化瘀方对原代培养CCl4纤维肝细胞胶原生成率及白蛋白(A1b)分泌功能的影响。方法:制备CCl4肝纤维化模型,胶原酶原位灌流法分离纤维肝细胞,以^3H-TdR掺入,^3H-Proline掺入胶原酶消化,ELISA等方法研究扶正化瘀方的血清药理学作用,结果:纤维肝细胞其增殖功能,A1b分泌功能的均较正常肝细胞低下,而胶原生成率则显著上升。
Aim: To investigate effects of FuZheng HuaYu decoction on cultured primary hepa-tocyte collagen synthesis and albumin secretion in carbon tetrachlorideinduced fibrotic rats with seropharmalogical method. Methods: The liver fibrotic model was induced by carbon tetrachloride in rats, then hepatocytes were isolated from models by in situ perfusion of liver with collagenase and were incubated with the serum from rats feed on FuZheng HuaYu decoction. The cells proliferation was measured by H-TdR incorporation, Cells collagen synthesis rate by H-Proline impulse and collagenases digestion and cells medium albumin by ELISA, and effects of the drug serum on fibrotic hepatocytes functions were observed. Results: Compared with normal hepatocytes , fibrotic hepatocytes producing collagens rate was higher, but cell proliferate and albumin secretion decreased. While the drug serum could inhibit fibrotic hepatocytes proliferation and collagens synthesis markedly, but improve their albumin production. Conclusion: Above effect can be one of main mechanisms of FuZheng HuaYu decoction anti-liver fibrosis actions, also reflect the supporting vital Qi and eliminating stasis actions of the decoction.
