多粘类芽孢杆菌Cp-S316抗真菌活性物质的提取及其部分性质研究 被引量:10

摘要 多粘类芽孢杆菌Cp-S316对动植物真菌、细菌病原物具有广谱抗性,为研究抗真菌活性物质的化学结构、理化性质并探讨其工业化生产的可行性,研究了大孔吸附树脂对多粘类孢杆菌Cp-S316产生的抗真菌活性物质的吸附、解吸性能,筛选出其最佳解吸剂,并测定了抗真菌活性物质的部分性质。结果表明,大孔吸附树脂X-5对抗真菌活性物质的吸附及解吸性能最好,其饱和吸附量为5.36×104μg·g^(-1),其最佳解吸剂为0.01mol·L^(-1)HCl-70%乙醇,以解吸剂∶树脂=2∶1(v∶m)进行静态解吸,解吸率达93.28%。该活性物质对热、紫外线稳定,对胰蛋白酶、蛋白酶K有一定的耐受性,对氯仿部分敏感,对酸稳定,对碱敏感。抗真菌活性物质提取方便、理化性质稳定,显示了一定的应用和开发前景。 Paenibacillus polymyxa Cp-S316 showed good antagonistic activity to many propagation pathogenic fungi and bacteria. To study the structure and physical-chemical characteristics of antifungal active substances produced by Paenibacillus Cp-S316, and also to probe into the feasibility of industrialization production, the absorption and desorption capacities of different macroporous adsorptive resins for active substances were researched. The macroporous adsorptive resin that had good absorption and desorption capacity was further investigated on desorption solution, and the optimum desorption solution was chosen. With the sample of crude antifungal active substances, some properties of active substances were characterized. The results showed that adsorption and desorption capacities of macroporous adsorptive resin X-5 for active substance was the best. The saturated absorption quantity of X-5 for active substance was 5.36×10^4μg·g^-1, and in the optimum des- orption solution, 0.01 mol· L^-1 HC1-70% ethanol desorbing in static state (desorption solution:resin=2:1 (re:V)), the desorption rate was up to 93.28%. The antifungal active substances indicated some good properties for application, such as they were stable to heat and uhraviolet ra- diation, not sensitive to trypsin, proteinase K and partial sensitive to chloroform, and also it showed higher activity in the solution with pH 4- 7, and did not indicate activity in alkali solution. Great prospect of application and exploitation was expected for their simple extraction and stable physical-chemical characteristics.
