摘要
目的:建立肝再生刺激物受体(HSSR)的荧光配体分析法,探讨该反应系统的最适反应条件,分析HSS与HSSR结合的特异性。方法:用己硫氰酸荧光素(FITC)标记HSS,通过直接荧光法观察。结果:肝细胞浓度为5×106/ml,FITCHSS1∶4稀释,4℃反应4h即可得到满意结果。结论:肝细胞膜上存在HSSR,对进一步探讨HSS作用的分子生物学机理具有重要意义。
Objective:To detect the hepatocyte stimulating substance receptors (HSSR) on fetus hepatocytes primary culture with fluoresceinligand binding assay.Method:HSS labelled with fluorescein isothiocyanate (FITC) was used for staining the hepatocytes.Results:Satisfactory results were observed with hepatocytes in concentration of 5×10 5/ml,FITC HSS in 1∶4 dilution and kept at 4℃ for 4 h.The HSSR was shown to be located on membrances of hepatocytes.Conclusion:This assay is important for studying the mechanism of HSS on hepatocytes.
