通心络对大鼠血管内皮早期损伤的保护作用 被引量:4

The Protective Function of Tongxinluo to Endothelium Injury by AngiotensinⅡOsmotic Pump in the Rat

摘要 目的:探讨通心络对大鼠损伤内皮是否有保护作用。方法:SD大鼠50只,分成假手术组(对照)10只、内皮损伤组20只和通心络组20只。每个组又分为1、3、7 d和14 d组4个时段。对照组动物仅在背部埋置0.9%氯化钠缓释泵;内皮损伤组在背部埋置血管紧张素Ⅱ缓释泵,剂量为200 ng·min^(-1)·kg^(-1),通心络组埋置血管紧张素Ⅱ缓释泵前3 d和处死前用通心络灌胃(剂量5 mg·kg^(-1)·d^(-1))。动物分别在术后1、3、7 d和14 d处死,检测血液粘度、透射电镜观察内皮形态的变化、比较肾脏组织中的ROS水平和P53mRNA水平。结果:内皮损伤组(血管紧张素Ⅱ组)见内皮细胞充血、水肿、脱落,内弹力板暴露、。而通心络组损伤较轻。通心络组可以降低肾组织中的活性氧自由基群(reactive oxygen spices,ROS)产生。同时还可以降低P53mRNA水平。在假手术组,上述指标无明显改变。结论:通心络具有降低血粘度,降低肾脏组织ROS水平和P53mRNA水平,对内皮细胞有一定的保护作用。 Objective:To explore whether Tongxinluo protect the endothelium injury induced angiotensin Ⅱ osmotic pump in rat. Methods:50 SD rats are divided into sham group( 10 rat ), endothelium injury group(20rat) and Tongxinluo group(20). There are four time points, that is, 1 d,3 d,7 d and 14 d in each group. Angiotensin Ⅱ osmotic pump at the dose 200 ng·min^-1·kg^-1 were embedded in the rat back of endothelium injury group,Physlological saline osmotic pump in the sham group. In treatment group, and Tongxinluo(5 mg·kg^-1·d^-1) was given by gastric tube after Angiotensin Ⅱ pump were used. All animals were sacrificed in 1^st, 3^rd ,7^th and 14^th day at the postoperation. The blood viscosity,reactive oxygen spices level and mRNA expression of P53 in the kidneys were examined by different methods. The morphology of endothelial cell of the renal artery were observed by transmission electron microscope. Results: the congestion, swelling, denudation of endothelial cell were seen in the endothelium injury group,but only light changes observed in Tongxinluo group. The level of reactive oxygen spices and P53 mRNA in Tongxinluo group was lower than that endothelium injury group. But Above indexes in sham group does not change apparently. Conclusion:Tongxinluo could protect endothelial cell and inhibit the production of reactive oxygen species. The medicine also could inhibit the cell apoptosis by lowering the level of P53 mRNA in the kidney.
