高效液相色谱法测定冬虫夏草中虫草素和虫草酸的含量 被引量:19

Determination of Cordycepin and Cordycep Acid in Cordyceps Sinensis by High-performance Liquid Chromatography

摘要 为建立检测冬虫夏草中虫草素和虫草酸的方法,用甲醇超声提取虫草素、水超声提取虫草酸,再用高效液相色谱、示差折光检测器与紫外检测器串联的方法测定虫草素和虫草酸含量。通过对样品进行预处理、色谱条件和方法学指标的验证,建立了定量检测虫草素、虫草酸的新方法,二者回收率在90%~101%之间,二者日内、日间精密度(RSD)≤7.2%。虫草素、虫草酸检出限分别为50ng/mL和1.5μg/mL。该方法具有快速、准确、简便的特点。 A high performance liquid chromatographic (HPLC) with diode-array detector (DAD) and refractive index detector (RID) method for the determination of cordycepin and cordycep acid in cordyceps sinensis was studied. The methods of extraction, preparation, chromatographic conditions and methodological index were tested and verified. An ideal method for the determination of cordycepin and cordycep acid in cordyceps sinensis was established. This method showed high speed and accuracy and was easy to perform. Ultrasonic with methanol was used for the extraction of cordy- cepin, and ultrasonic with water was used for cordycep acid extraction. Both the mobile phase for analysis cordycepin and cordycep acid were acetonitrile and water. Their recovery were between 90% and 101%, their precision (RSD) within the day and between the days were lower than 7.2%.
