摘要
目的通过检测基质溶解因子(matrilysin,MMP-7)在人直肠癌中的表达,探讨其在直肠癌浸润与转移中的作用。方法对100例直肠癌与自身配对的正常直肠组织采用实时荧光定量逆转录-多聚酶链反应(Quantitative-Real-Time RT-PCR)、免疫组化及计算机图像分析方法检测MMP-7mRNA和蛋白水平的表达,定量分析其表达与临床病理特征的关系。结果MMP-7在肿瘤组的mRNA表达水平高于正常组(P=0.001),表达比值大于1的67例(67%),mRNA表达与Dukes分期、淋巴结转移及组织学分化程度相关。MMP-7在直肠癌中的免疫组织化学染色强度(1.94±0.21)高于正常组(1.15±0.20,P=0.002)。蛋白表达与年龄、Dukes分期及淋巴结转移相关。结论MMP-7在直肠癌中的表达明显升高,在直肠癌的浸润与转移中具有重要作用。
Objective To detect the matrilysin (MMP-7) expression in human rectal cancer and to investigate whether it is correlated with invasion and metastasis of human rectal cancer. Methods The paired samples obtained from 100 inpatients were allocated into cancer group and control group. By Quantitative-Real- Time RT-PCR, immunohistoehemieal staining and computerized image analysis, the mRNA and protein expressions of MMP-7 in human rectal cancer were measured and further analyzed for the relationship between MMP-7 expression and elinieopathologie characters. Results MMP-7 mRNA expression in cancer group was higher than that in control group (P=0.001), the mRNA expression ratio of 67 (67%) samples was over 1. The mRNA expression level of MMP-7 was correlated with age, Dukes's Staging, lymph node metastasis and histological differentiation grade. The positive degree of immunohistoehemieal staining for MMP-7 in cancer group (1.94±0. 21) was higher than that in control group(1.15±0.20, P=0.002). The protein expression of MMP-7 was correlated with age, Dukes's Staging and lymph node metastasis. Conclusion MMP-7 expression in human rectal cancer increases significantly and plays a key role in the invasion and metastasis of human rectal cancer.
