摘要
目的进行小鼠甲状腺球蛋白的纯化和鉴定。方法小鼠甲状腺组织匀浆后进行盐析和Sphadex G-200凝胶层析2次纯化,纯化后的蛋白质产物经十二烷基磺酸钠—聚丙烯酰胺凝胶电泳(SDS-PAGE)和Western免疫印迹鉴定。结果经纯化的蛋白产物为高纯度的甲状腺球蛋白。结论Sphadex G-200凝胶层析和Western免疫印迹是进行甲状腺球蛋白纯化和鉴定的一种较好的方法。
[Objective] To purify and identify thyroglobulin of mouse. [Method] Thyroid gland tissues were homogenized and salted out, and then purified by repeated gel filtration on Sphadex G-200. This protein was identified by SDS-PAGE and western Immunoblotting decting. [Result] This protein was verified as the high purified Thyroglobulin. [ Conclusion] Sephadex G-200, together with Immunoblotting is a good way to purify and identify thyroglobulin of mouse.
