The Association of MMP-1-1607(1G-2G) Single Nucleotide Polymorphism with the Susceptibility to Non-small Cell Lung Carcinoma in Guizhou Han Nationality

摘要 目的:研究基质金属蛋白酶1(metal matrix proteinase-1,MMP-1)启动子区-1607bp处1G或2G单核苷酸多态性(single nucleotide epolymorphism,SNP)与贵州汉族人非小细胞肺癌(non—small cell lung carcinoma,NSCLC)遗传易感性的关系。方法:采用聚合酶链反应-限制性片段长度多态性(polymerase chain reaction—restriction fragment length polymorphism,PCR—RFLP)方法分析71名NSCLC患者和75名健康对照者的MMP-1启动子区SNP的基因型,比较不同基因型与NSCLC发生风险的关系。结果:2G/2G基因型在NSCLC组中分布频率高于健康对照组。2G/2G基因型较1G/1G及2G/2G+1G/2G基因型患NSCLC风险率增加,OR(95%CI)值分别为3.082(1.620—5.862),2.517(1.283—4.936),吸烟可增加2G/2G基因型患NSCLC风险率。结论:贵州汉族人群MMP-1启动子区SNP(1G/2G)基因型与NSCLC易感性有关,2G/2G基因型的NSCLC发生风险增加。 Objective: To investigate the relation of the single nueleotide polymorphism (SNP) in matrix metalloproteinase 1 (MMP-1) gene with the susceptibility to non-small cell lung carcinoma (NSCLC) of Han nationality in Guizhou province. Methods: MMP-1-1607 (1 G-2G) of 71 NSCLC patients and 75 healthy people were amplified with polymerase chain reaction-restriction fragment length polymorphism analysis (PCR-RFLP) technique, and the polymorphism was analyzed. The relationship of different genotypes with the susceptibility of NSCLC was analyzed. Results: The distribution frequency of individuals with genotype 2G/2G was significantly higher in NSCLC patients than that in healthy controls. The individuals with genotype 2G/2G had higher risk in suffering from NSCLC than individuals with genotypes of 1G/2G or 2G/2G + 1G/2G, and the OR values (95% CI) were 3. 082 ( 1. 620 -5. 862) and 2.517 ( 1. 283 -4. 936) respectively. Smoking could increase the risk of NSCLC. Conclusions: SNP of genotype (1G/2G) in promotor area of MMP-lgene in Han people of Guizhou relates to the susceptibility of NSCLC. People have genotype 2G/2G have higher risk to be attacked by NSCLC.
