摘要 目的:测定华蟾素(cinobufacine,Cino)对耐阿霉素人乳腺癌细胞MCF-7/ADM内阿霉素(ADM)积累的影响,以探索Cino逆转MCF-7/ADM多药耐药(MDR)的可能机制。方法:MTT法检测CinoMDR的逆转作用,HPLC法检测Cino作用MCF-7/ADM后细胞内ADM的浓度的变化。结果:Cino15mg/L能增加MCF-7/ADM细胞对ADM的敏感性,使ADM的半数抑制浓度(IC50)由38.14mg/L降至12.93mg/L,显著增加MCF-7/ADM株细胞内ADM的浓度。结论:Cino能明显增加MCF-7/ADM株细胞内ADM的含量,部分逆转MCF-7/ADM的MDR。 AIM: To study the effects of cinobufacine on intracellular adriamycin accumulation in human breast cancer MCF-7/ADM cells, and further explore the mechanism of cinobufacine reversing the multidrug resistance of human breast cancer MCF-7/ADM cells. METHODS: The cytotoxic effect was determined by MTT assay. Intracellular ADM concentration was examined by HPLC. RESULTS: 15 mg/L cinobufacine decreased the IC50 of ADM to MCF-7/ADM cells from 38.14 mg/L to 12.93 mg/L, but increased intracellular adriamycin accumulation in MCF-7/ADM cells. CONCLUSION: Cinobufacine can increase intracellular ADM accumulation and partly reverse MDR of MCF-7/ADM cell line.
