摘要
胰岛素刺激引起的葡萄糖转运主要由葡萄糖转运子(GLUT)4介导。GLUT4的亚细胞分布、基因表达量以及功能活性,都将直接降影响胰岛素作用下葡萄糖的跨膜转运。高葡萄糖和高胰岛素可加强己糖胺途径来抑制GLUT4转位过程、下调胰胰岛素受体底物的蛋白水平和酪氨酸磷酸化、从转录和转录后水平抑制GLUT4的基因表达、直接降低GLUT4内在活性,继而使胰岛素通过GLUT4介导的糖转运下降,引起胰岛素抵抗。
Insulin-induced glucose transportion is mainly mediated by GLUT4. The subcellular distribution, gene expression, and inherent activity of GLUT4 directly affect the glucose transmembrane transportation induced by insulin. By strengthening hexosamine pathway, high glucose and hyperinsulinism can achieve some effects in inhibiting transposition of GLUT4, down-regulating protein protein level and tyrosine phosphorylation of IRS, depressing gene expression of GLUT4 on transcriptional and post-transcriptional level, directly reducing inherent activity of GLUT4 and then decreasing GLUT4-induced glucose transportation of insulin, which then result in insulin resistance.
