摘要
本文根据系统分析方法,利用正交旋转组合设计于固体培养系统中测定培养基的装样量、细菌培养时间、接线虫量和线虫培养时间对Steinernemacarpocapsae和Heterorhabditisbacteriophora产量的综合影响,确定线虫的优化培养参数。在设计组合范围内,接线虫量对A24线虫产量的影响显著;培养基的量和细菌培养时间对线虫产量的影响不显著。对H06线虫产量,接线虫量、培养时间呈显著的正影响;细菌培养时间则呈显著的负影响。此外,接线虫量与培养时间对两种线虫均产生明显的负交互效应。培养基的装样量对线虫产量无显著影响。通过频数分析,分别得到了A24和H06线虫的培养参数的优化组合。即每克培养基的A24线虫产量大于50×104时,培养参数的优化组合是培养基的量为每瓶53.6~58.8克,细菌培养时间2天左右,接线虫量约每克培养基4.6×103,培养时间为16~18天。同理,每克培养基的H06线虫产量大于25×104时,培养参数的优化组合分别为每瓶培养基的量67.8~74.2克,细菌培养时间1~2天,接线虫量为每克培养基5.5×102~2.2×103之间,培养时间22~24天。根据这些培养参数的优化?
Effects or interactions of four culture parameters on the yield of Steinernema carpocapsae A24 and Heterorhabditis bacteriophora H06 were determined in solid cultures. Yields of infectives were significantly positively influenced by nematode inoculum size(ranging from 4.6 ̄4.6×10 4 for A24 and 5.5 ̄5.5×10 4 for H06 per g medium) and culture time of nematodes (ranging from 8 ̄24 days for A24 and 10 ̄30 days for H06) for both strains, and significantly negatively influenced by bacterial culture time(0 ̄4 days)for H06, but not by quantity of medium per flask (20 ̄100g/flask) for both strains or by bacterial culture time for A24. There were significant negative interactions between nematode inoculum size and culture time of nematodes on both strains. Optimum culture parameters were modeled for both strains and commercial operation in the culture system was suggested
