海生素对体外白血病K562细胞的抑制作用及其机制 被引量:2

摘要 目的探讨海生素对体外白血病K562细胞的抑制作用及机制。方法将0.1、1、10、100、1000μg/ml的海生素分别作用于K562细胞24、48 h,免疫细胞化学法检测凋亡相关蛋白bcl-2、bax的表达。结果海生素作用后K562细胞抑制率明显高于对照组,且呈现浓度及时间依赖性。K562细胞bcl-2蛋白表达下调,bax蛋白表达上调。结论海生素可抑制K562细胞的生长,其机制可能为通过下调bcl-2蛋白表达,增加bax蛋白的表达,诱导细胞凋亡。 [Objective]To study the effect of Haishengsu on bcl-2 and bax protein expression and K562/ADM cells in vitro. [Methods] MTT assay was used to determine the effect of haishengsu on K562 cells,immunocytochemistry was used to measure the expression of bcl-2 and bax. [Results] Haishengsu inhibited the proliferation of K562 cells,Haishengsu down-regulated the expression of bcl-2 and upregulatea the expression of bax in K562 cells. [Conclusion] Haishengsu can inhibit the proliferation and induce apoptosis of K562 cells by downregulating bcl-2 expression and up-regulating bax expression.
