摘要
为探讨卵泡刺激(FSH)与黄体生成生素(LH)分泌及基因表达与调控用抗人LH-β亚单位单克隆抗体和抗人FSH-β亚单位单克隆抗体进行免疫细胞化学定位的研究。通过连续切片免疫酶技术,免疫细胞化学双标记及电镜免疫金双标记技术,在大鼠垂体前叶进行FSH和LH的免疫细胞化学共定位研究。结果表明:存在着棕(DAB显色示FSH)、蓝(CN显色示LH)及棕蓝混合染色细胞;电镜免疫金双标记发现分别被15nm金粒(示FSH)、10nm(示LH)及被15nm、10nm金粒同时标记的3种细胞(另文报道);光镜连续切片染色亦发现FSH阳性、LH阳性及FSH-LH双阳性3种细胞。因此可以确定垂体前叶存在3种促性腺激素(GTH)细胞,即FSH细胞(只含FSH)、LH细胞(只含LH)和FSH-LH细胞(既含FSH又含LH)。
The cellular co localization of follicle stimulating hormone (FSH) and luteinizing hormone (LH)in the pituitary gland applying the anti β FSH/β LH McAb through the techniques of immunoenzyme on the serial paraffin sections and immunocytochemistry with double labelling had been proceeded.The results showed that there was a real co localization of FSH and LH on gonadotrophs: Three different kinds of cells could be identified by colour difference of immunostaining,these were brown ( FSH cell only).blue(LH cell only) and bluish brown (FSH LH cell) after utilizing of immuno double labelling. Meanwhile, three kinds of cells (only FSH positive staining cells, only LH positive staining cells and both FSH and LH positive staining cells) were distinguished after immunostaining on serial sections. Therefore, the conclution was obtained that there were three kinds of gonadotrophs in pituitary: FSH cells,LH cells and FSH LH cells. The possible reasons why exist these three types gene expression of gonadotrophic hormones in the anterior pituitary were discussed.
