肝X受体(LXR)是与代谢综合征相关的核受体之一,对脂代谢的转录调控起重要作用。LXR是多种细胞内胆固醇含量的感受器,被激活后,诱导一系列与胆固醇吸收、转运和分泌相关的基因表达,并促进脂肪酸和甘油三酯的合成。LXR激动剂减少肝糖异生,降低血浆葡萄糖水平,增加胰岛素抵抗动物模型的胰岛素敏感性。LXR是胰岛素信号通路的组成部分,与能量平衡和肥胖相关,并参与调节炎性反应和氧化应激反应。作为代谢通路与炎性反应信号通路的结合点,LXR可能成为代谢性疾病治疗的新靶点。
The liver X receptor (LXR) is one of the nuclear receptors related to metabolic syndrome. LXR plays an important role in the transcriptional control of lipid metabolism and acts as a nuclear cholesterol sensor in multiple cell types. Once activated, LXR induces the expression of an array of genes involved in cholesterol absorption, transport, and excretion, as well as fatty acids and triglyceride synthesis. In addition, synthetic LXR agonists have been found to reduce glyeoneogenesis in liver, decrease plasma glucose level and improve instdin sensitivity in insulin resistance animal models. LXR is one part of instdin signaling, which is related to obesity and energy balance, and also a modulator of inflammatory response and oxidative stress. The ability of LXR to integrate metabolic and inflammatory signaling makes it particularly attractive target for intervention in human metabolic diseases.
