摘要
目的:探讨IL6信号传递系统中GP130,JunB是否参与了弥漫性增生性狼疮性肾炎的分子病理过程。方法:采用原位杂交技术观察了20例弥漫性增生性狼疮性肾炎患者肾活检组织肾小球细胞中IL6、GP130、JunBmRNA表达,并分析其与患者肾小球系膜区增宽,尿蛋白之间的关系。结果:弥漫性增生性狼疮性肾炎患者IL6、GP130、JunB基因mRNA表达分别是66±09,86±09,67±08,较对照组明显增加(Ρ<001);IL6、GP130mRNA表达与系膜区增宽、尿蛋白排泄量呈正相关。结论:弥漫性增生性狼疮性肾炎IL6及其信息传递中的GP130,JunB基因异常表达在其分子发病机理起一定作用。
OBJECTIVE To observe the expression of IL 6, GP130 and Jun B genes in the renal tissues of patients with diffuse proliferative lupus nephritis(LN). METHODOLOGY The expression of the target mRNAs in glomeruli were detected in 12 patient with active LN by using in situ hybridization technique. with digoxigenin labeled probes. Semi quantitative technique was applied by scanning the signals of mRNA in unit glomerular area. RESULTS The expression of IL 6, GP130 and Jun B genes in glomeurli of LN patients (6 6±0 9, 8 6±0 9 and 6 7±0 8, respectively) were significantly higher than that of the normal control( P <0 01 for all); and positive correlations were found between each other. The IL 6 and GP130 mRNA levels were found positively correlated with the mesangial expansion and urinary protein excretion respectively. While Jun B mRNA level was not associated with mesangial expansion or urinary protein excretion. CONCLUSION Abnormal gene expressions of IL 6, Jun B and GP130 might participate in the molecular pathogenesis of lupus nephritis.
