摘要
目的探讨超声微泡造影剂介导4-羟基他莫昔芬(4-hydroxytamoxifen,OHT)治疗大鼠前列腺增生的疗效。方法分别用超声微泡(UM)、OHT、UM+OHT治疗前列腺增生大鼠,测其前列腺指数;光镜下观察前列腺细胞形态;免疫组化测定前列腺组织中Bax和Bcl-2的定位表达,并利用图像分析技术作定量分析。结果UM+OHT治疗组与正常对照组前列腺指数值较阳性对照组、OHT治疗组及UM组明显减小(P〈0.05),阳性对照组与UM组腺体重度增生,OHT组中度增生,UM组轻度增生或无增生,与正常对照组相似。Bax与Bcl-2在前列腺内分布基本一致,在前列腺上皮和基质均有表达,以上皮细胞为主,呈胞浆、胞膜型。Bcl-2在阳性对照组、UM组表达最高,UM+OHT治疗组与正常对照组表达最低;Bax在阳性对照组、UM组表达下降,正常对照组、OHT及UM+OHT组表达升高。结论抗雌激素药物OHT对大鼠实验性前列腺增生有一定治疗作用,超声微泡与OHT联合作用时可使OHT疗效增加。
Objective To investigate the treatment of ultrasound microbubble (UM) mediated 4- hydroxytamoxifen(OHT) in rat with benign prostate hyperplasia(BPH). Methods Fifty rats with BPH were respectively treated with UM,OHT and UM + OHT, prostatic index(H) was assessed and the morphology of prostatic tissue was observed under microscope. The localization expression of Bax and Bcl-2 in prostatic tissue was determined by immunohistochemistry and quantitative analysis by using image analysis. Results PI was significantly decreased in the control group and UM + OHT group than in BPH group,OHT group and UM group ( P 〈0. 05). The hyperplasia of prostatic tissue was the most in BPH and UM group,moderate in OHT group, minimal in the control and UM + OHT group. The distribution of expression of Bax and Bcl-2 in the prostatic tissue was basically identical,which were mainly expressed in prostatic epithelium,as well as matrix, the Bax and Bcl-2 presented in cytoplasmic and membrane. The expression of Bcl-2 was the highest in BPH and UM group, lowerest in the control and UM + OHT group. The expression of Bax was decreased in BPH and UM group, increased in the other three groups. Conclusions OHT has therapeutically effect on rat with BPH and the effect was increased by using of UM. UM can also downregulate the expression of Bcl-2 and lead to increase of Bax, thereby induce apoptosis of prostatic cell.
