摘要
【目的】建立高效液相色谱(HPLC)法测定大鼠血浆中五味子甲素的方法。【方法】以甲醇沉淀蛋白,色谱柱为C18柱(Intersil:150 mm×4.6 mm,5μm),流动相为甲醇-乙腈-水(42∶40∶18),流速为1.0ml/min,检测波长为λ=254 nm。【结果】五味子甲素在0.1~5μg/ml范围内有良好的线性关系:Y=0.3047C+0.0315,r=0.9975(n=5)。五味子甲素的方法平均回收率为95.36%,RSD为4.48。日内精密度RSD为1.99%。日间精密度RSD为2.87%。【结论】首次建立了测定五味子甲素血药浓度方法,该方法简便、快速,适于测定鼠血浆中的五味子甲素。
[Objective] To estabhsh a HPLC method for determination of deoxyschizandrin in rat's plasma. [Methods] The analytical column was C18 (Intersil: 150 mm × 4.6 mm, 5μm) with a mobile phase of methanol-acetonitrile-water (42:40:18). The flow rate and detection wave length were 1.0 ml/min and 254 nm respectively. [Results] The calibration curve of deoxyschizandrin was hnear within the rang of 0.1 ~ 5 g/ml, and the regression equation was Y = 0. 3047C + 0.0315, r = 0. 9975 ( n = 5). The average recovery of deoxyschizandrin was 95.36%, RSD 4.48%. The average RSD of intra-day and inter-day validation were 3.57%and 1.99% respectively. [Conclusions] For the first time we have established a method to measure the blood levels of deoxyschizandrin easily and quickly, which may be apphed to the measurement of deoxyschizandrin in rat's plasma.
