摘要
目的:探讨改善微循环对周围神经嵌压性损害血管内皮细胞生长因子(VEGF)表达和病理改变的影响。方法:分别检测坐骨神经嵌压后12、72h和7d背根神经节细胞VEGF水平与第4周神经干、神经节病理变化、脊髓病变神经元计数等,并运用正常对照组、模型组、前列地尔组和丁咯地尔组采用SPSS10.0统计软件进行方差分析统计处理。结果:嵌压后72hVEGF水平达到高峰,与正常对照组比较,其余各组均显著增加(P<0.01,P<0.05);前列地尔组和丁咯地尔组较模型组为优(P<0.01,P<0.05);嵌压后4周前列地尔组和丁咯地尔组的神经干、神经节病理变化、脊髓病变神经元计数均优于模型组(P<0.05)。结论:对于周围神经嵌压性损害,改善微循环可增加VEGF的表达并减轻其病理损害,从而对周围神经嵌压性损害的修复具有促进作用。
Aim: To study the influences on expression of vascular endothelium growth factor (VEGF) and pathologic changes by improving microcirculation following crushing lesion of sciatic nerve in rats. Methods: Sciatic nerve was crushed by pincers to establish the model of sciatic nerve crush lesion in rats. The expression of VEGF in dorsal ganglion cells at 12 h, 72 h, 7 d and the pathologic changes of sciatic nerve, dorsal ganglion and spinal cord at 4th week were examined respectively by immunohistochemical and pathological techniques, then their results were compared each other. SD rats were divided into normal group, model group, alprostadil (Lipo-PGE0 group and buflomedil hydrochloride group at random. There were 16 rats in each group. SPSS 10.0 was used statistically. Results: The expression of VEGF in dorsal ganglion cells increased from 12 h after crush injury of the nerve, and have increased to the highest from 72 h after crush injury of the nerve (P 〈 0.01). There was no increasing of it in normal group. It increased in alprostadil group and buflomedil hydrochloride group more than model group significantly(P 〈 0.01). It became normal on 1st week in all groups after sciatic nerve crush injury. On 4th week after sciatic nerve crush injury the severity of pathologic outcomes (including spinal neuron counting) in alprostadil group and buflomedil hydrochloride group was slighter than it in model group, while there was no pathologic change in normal group. Spinal neurons counting of alprostadil group and buflomedil hydrochloride group were more optimal than those of model group significantly (P 〈 0.01). Conclusion : The expression of VEGF increases and the pathologic changes of sciatic nerve become slight by improving microcirculation after peripheral nerve crush lesion, so that microcirculation has improving action to repair peripheral nerve crush lesion.
