心肌神经生长因子在心肌梗死后的动态表达

摘要 目的:观察心肌梗死后大鼠心肌中神经生长因子蛋白及mRNA表达的动态演变过程。方法:实验于2004-10/2005-04在哈尔滨医科大学附属第一医院中心实验室完成。①实验动物:8~12周龄Wistar雄性大鼠68只。②实验分组:53只大鼠存活。其中9只为3d心肌梗死组,10只为7d心肌梗死组,10只为30d心肌梗死组,其余24只大鼠均分为各组的假手术组。③实验干预:参照Kin等方法制作大鼠的左室心肌梗死模型,假手术组为穿线后不结扎冠状动脉。④实验评估:术后3,7,30d从各组大鼠梗死中心、梗死周边、室间隔和右室取材,通过原位杂交及免疫组化方法检测神经生长因子mRNA及蛋白的表达。结果:53只大鼠进入结果分析。①心肌梗死后3d,梗死中心区出现一过性的神经生长因子蛋白及mRNA的阳性表达,神经生长因子蛋白的表达在梗死周边心肌中明显减少(t=2.142,P<0.05),在右心室中增加(t=3.045,P<0.01)。②心肌梗死后7d,梗死周边、室间隔、右心室中神经生长因子蛋白的表达明显高于假手术组(t=2.347,P<0.05;t=3.834,P<0.01;t=2.979,P<0.01);室间隔中神经生长因子mRNA的表达明显高于心肌梗死后3d组(P<0.01)。③心肌梗死后30d,神经生长因子蛋白的表达在右室中明显高于假手术组(t=2.056,P<0.05);而室间隔中神经生长因子mRNA的表达明显高于假手术组及心肌梗死后3d(t=3.425,P<0.01;t=3.063,P<0.01)。结论:心肌梗死后不同时间点,不同部位心肌神经生长因子mRNA及蛋白的表达不同,提示神经生长因子可能在心肌局部发挥神经营养作用进而参与神经重构及神经内分泌的调节过程。 AIM: To explore the dynamic process of the expression of nerve growth factor (NGF) in myocardium of rats after myocardial infarction. METHODS: The experiment was conducted in the central laboratory of First Affiliated Hospital of Harbin Medical University from October 2004 to April 2005. ①Sixty-eight male Wistar rats (8-12 weeks old) were selected and only 53 survived after operation. They were divided into 3 (n =9), 7 (n =10), 30 (n =10) days infarcted groups and sham operation group (n =24) as control. ②According to Kin's method, rat model of left ventricular infarction was made, and the left coronary artery of sham operation group was not ligated. In infarcted zone, peri-infarcted area, septum and right ventricle, the expressions of NGF protein and mRNA were examined by in situ hybridization and immunohistochemistry 3, 7, and 30 days after operation. RESULTS: A total of 53 rats were involved in the result analysis. ①In 3 days infracted group, the expressions of NGF mRNA and protein were detected in infarcted cardiomyocytes, and the expression of NGF protein was significantly lower in peri-infarcted area (t =2.142, P 〈 0.05) and higher in right ventricle than control (t =3.045, P 〈 0.01). ②In 7 days infracted group, the expression of NGF protein in peri-infarcted area, septum and right ventricle were significantly increased compared with that in sham operation group (t =2.347, P 〈 0.05; t =3.834, P 〈 0.01; t =2.979, P 〈 0.01), and the expression of NGF mRNA in septum was significantly higher than that in 3 days infarcted group (P 〈 0.01). ③In 30 days infarcted group, the expression of NGF protein in right ventricle was higher than that in sham operation group (t =2.056, P 〈 0.05), and the expression of NGF mRNA in septum was higher than sham operation group and 3 days infracted group (t =3.425, P〈 0.01; t =3.063, P〈 0.01). CONCLUSION: The expressions of nerve growth factor protein and mRNA in myocardium at different time points and sites dynamically change, which indicates that nerve growth factor may play a nerve-nutritious role and take part in the post-infarcted nerve remodeling and neuroendocrine regulation.
