乌司他丁联合三七总皂甙对急性胰腺炎大鼠氧自由基的影响 被引量:9

摘要 目的:探讨乌司他丁、三七总皂甙合用对急性胰腺炎(AP)大鼠氧自由基的影响.方法:SD大鼠75只,随机分为A,B,C,D,E5组.A组为假手术组;B组为急性胰腺炎组;C组为乌司他丁治疗组;D组为三七总皂甙治疗组;E组为乌司他丁联合三七总皂甙治疗组.通过胰胆管逆行性注射牛磺胆酸钠制成AP大鼠模型.分别采用黄嘌呤氧化酶法和硫代巴比妥酸法于术后3 h,6 h,12 h测定各组大鼠血清超氧化物歧化酶(SOD)和丙二醛(MDA)浓度.结果:B组与A组相比大鼠表现为血清SOD降低和MDA升高.术后6 h,C组和D组较B组血清SOD明显升高(274.750±14.44 U/L,281.420±16.48 U/L vs 204.440±14.87 U/L,P<0.05),MDA明显降低(6.85±0.65μmol/L,6.78±0.56μmol/L vs 9.25±0.22μmol/L,P<0.05);E组较C组血清SOD明显升高(318.360±13.71 U/L vs 274.750±14.44 U/L,P<0.05),MDA明显降低(5.53±0.45μmol/L vs 6.85±0.65μmol/L,P<0.05);C组与D组相比没有显著差异.结论:乌司他丁、三七总皂甙通过升高SOD保护性因子降低MDA损伤性因子,对AP起治疗作用.而乌司他丁联合三七总皂甙应用具有协同效应. AIM: To investigate the effects of ulinastatin combined with panax notoginseng saponins (PNS) on oxygen free radicals in rats with acute pancreatitis (AP). METHODS: Seventy-five Sprague-Dawley rats were divided into five equal groups: group A (sham-operated), B (AP), C (treated with ulinastatin), D (treated with panax notoginseng sapo- nins) and E (treated with ulinastatin combined with panax notoginseng saponins). A rat model of AP was created by retrogression injection of sodium taurocholate though pancreas biliferous ducts, Serum superoxide dismutase (SOD) and malondialdehyde (MDA) levels in each experimental group were observed at 3, 6 and 12 h. RESULTS: The level of SOD in group B de- creased while the MDA increased. Levels of SOD in groups C and D were higher than that in group B (274.750 ± 14.44 U/L and 281.420 ± 16,48 U/L vs 204.440 ± 14.87 U/L, P 〈 0.05) while the levels of MDA were lower (6.85 ± 0.65 μmol/L and 6.78 ± 0.56 ~mol/L vs 9.25 ± 0.22 μmol/L, P 〈 0.05). The level of SOD in group E was higher than that in group C (318,360 ± 13.71 U/L vs 274.750 ± 14.44 U/L, P 〈 0.05) while the level of MDA was lower (5.53 ± 0.45 μmol/L vs 6.85 ± 0.65 μmol/L, P 〈 0.05). CONCLUSION: Ulinastatin in combination with PNS has therapeutic effects on AP by increasing the protective SOD and decreasing the MDA injury factors. The effect is more evident when ulinastatin is combined with PNS than when applied separately.
