摘要
本文从进境的美国大豆豆杆中分离到一种真菌,通过形态学特征鉴定、致病性检测和序列分析,结果为大豆北方茎溃疡病菌(Diaporthe phaseolorumvar.caulivora)。该菌子囊壳在豆杆上球形,黑色,常2~12个簇生,偶有单生,大小180~316μm×290~398μm;子囊壳具长喙,喙长251.1~1085.4μm,喙基部宽50.6-123.5μm;子囊长棍棒状,无柄,内含8个子囊孢子,大小为29.8~31μm×5~6.5μm;子囊孢子无色,长椭圆形,1隔,分隔处稍缢缩,两室两油球,10.1~11.3μm×2.43~3.24μm。菌落白色,后期变为白-黄色,菌丝紧密收缩,后逐渐变为茶色并蓬松。用ITS区通用引物对该菌DNA进行扩增和测序,获得522bp的ITS区序列。
Northern stem canker Diaporthe phaseolorum var. caulivora was identified from imported soybean stem from USA.The methods of morphology,pathogenicity and sequencing were used in this study. The perithecia on stems are black,globose ,occur in caespitose groups of 2~12,occasionally single perithecium.The perithecial dimensions are 180~316μm×290~398μm.The protruding perithecial beaks in length are 251.1~1085.4μm, at the base of beaks in width are 50.6~123.5μm. The 8-spored asci are sessile, elongate -clavate, the asci measured 29.8~31μm×5~6.5μm.The ascospores are elongate-ellipsoidial, bicellular, slightly constricted at the septum,and possess 2 guttulates per cell, the measurement are 10.1~11.3μm×2.43~3.24μm. The colonies are white-yellow, with compact, shrunken mycelia that change to ochre and fluffy with age. The ITS sequence of the fungus was obtained by PCR and sequenced,the result was D.p. var. caulivora.
