STUDY ON PROTECTIVE EFFECTS OF HO-1 AGAINST RAT RENAL CELL APOPTOSIS CAUSED BY MERCURY

摘要 [目的]探讨血红素氧合酶-1(HO-1)对汞引起肾细胞凋亡的影响及其可能的机制。[方法]64只雄性Wistar大鼠随机均分为4组:对照组、单纯染汞组、HO-1诱导组、HO-1抑制组,先分别腹腔注射生理盐水、空白液、血晶素、ZnPPⅨ预处理,8h后各组内半数鼠处死取肾查HO-1表达与活性,余鼠除对照组注射生理盐水外,其余3组均腹腔注射HgCl2(2mg·kg-1),24h后检测血Cr、BUN及肾内总抗氧化能力(TAOC)、丙二醛(MDA)、Bcl-2蛋白表达与细胞凋亡指数(AI)。[结果]与对照组比较,单纯染汞组TAOC降低而MDA、Cr、BUN、Bcl-2表达和AI均明显增加,HO-1抑制组除Bcl-2表达受抑外,其余指标的变化与单纯染汞组相似,但更为显著。与单纯染汞组及HO-1抑制组比较,HO-1诱导组TAOC及Bcl-2表达均显著升高,而MDA、Cr、BUN及AI则明显降低。[结论]HO-1通过抗氧化、上调Bcl-2蛋白表达发挥抗汞引起肾细胞凋亡的作用。 [ Objoctive] To investigate the effects of heine oxygenase-1 (HO-1) on rat renal cell apoptosis caused by mercury and the possible protective mechanisms. [Methods] 64 male Wistar rats were randomly divided into four groups: control group, simple mercury group, HO-1 induced group and HO-1 inhibited group. Rats in each group were intraperitoneally injected with normal saline, vehicle, heroin (30 mg.kg^-1) . ZnPPIX (20 mg,kg^-1) respectively. 8 hours later, half of rats in each group were killed and their kidneys were harvested for detecting the expression and activity of HO-1 protein. The other rats except the control group were intraperitoneally injected with 2 rng.kg^-1 mercuric chloride (HgCl2), 24 hours later, blood creatinine (Cr), blood urea nitrogen (BUN), and renal total antioxidative capacity (TAOC), malondialdehyde (MDA), Bcl-2 protein expression and cell apoptosis index (AI) were measured. [Results] Compared with the control group, the TAOC decreased, while the levels of MDA, Cr, BUN, Bcl-2 protein expression, and AI increased in simple mercury group. The expression level of Bcl-2 protein was inhibited in HO-1 inhibited group, besides, the changes of other parameters were just like but more obvious than those in simple mercury group. Otherwise. compared with simple mercury group, the levels of TAOC and Bcl-2 protein expressmn increased obviously, while the levels of MDA, Cr, BUN and AI decreased significantly in HO-1 induced group. [ Conclusions] HO-1 could protect against renal cell apoptosis caused by mercury by means of anti-oxidation and up regulation of Bcl-2 protein expression.
