摘要
目的测定眼镜蛇毒细胞毒素(CV-CTX)对人癌细胞株的细胞抑制作用,以期能简便准确地反映其抗癌活性。方法应用溴化二苯四偶氮盐(MTT)法测定在CV-CTX作用下体外培养的人胃癌细胞株(SGC-7901)和人肝癌细胞株(SMC)的存活情况和其量效关系。结果(1)SMC、SGC-7901的活细胞数与其分解MTT的量成正线性关系。(2)CV-CTX对SMC、SGC-7901癌细胞株的细胞抑制作用呈良好的量效关系,IC50分别为2.45和1.24μg/ml。结论(1)用MTT法测定癌细胞的增殖和衰减是可靠的。(2)应用MTT法证实CV-CTX对体外培养的人癌细胞的杀伤程度可作为其抗癌活性指标。
Objective\ CV CTX is a cytotoxin separated from CV (cobra venom,CV).\ To reflect precisely the anticancer activity of CV CTX on human cancer cell lines.\ Methods\ To study the effect of CV CTX on the survival rate of cultured human cancer cells and dose effect relationship by 3 (4,5 dimethylthiazol 2 yl) 2,5 diphenyl tetrazolium bromide(MTT) assay method.\ \ Results\ (1)The amount of living cultured cancer cells was well correlated with the amount of decomposed MTT.(2)The inhibition of CV CTX on SMC and SGC 7901 cell lines showed good dose effect relationship,the IC 50 was 2.45 and 1.24 μg/ml respectively.\ Conclusion \ (1)The MTT assay is a trust worthy method for testing the proliferation and attenuation of cancer cells.(2)The killing rate of CV CTX on the cultured cancer cell lines by using MTT assay was validated as an index of anticancer activity.
