摘要
目的:观察青刺果多糖对四氧嘧啶诱导的糖尿病小鼠肝组织病理变化的影响,并探讨其对糖尿病小鼠肝脏保护作用。方法:腹腔一次性注射四氧嘧啶复制模型。造模成功后,将实验动物分为模型组、青刺果多糖治疗组、阳性药物治疗组。另设正常对照组。实验小鼠分别于给药2周末和4周末处死,取肝脏称重,测定肝脏脏器指数,然后固定于4%的多聚甲醛中,制作病理切片,HE染色,光镜下观察。结果:青刺果多糖治疗组和阳性药物治疗组可显著降低糖尿病小鼠的肝脏脏器指数(P<0.05)。病理组织观察:模型组小鼠肝细胞索排列紊乱;肝血窦增宽,肝细胞索间纤维细胞增生,肝细胞发生脂肪变性和局灶性坏死,坏死灶内大量炎性细胞浸润,中央静脉周围炎性细胞浸润。经青刺果多糖治疗后,肝细胞索状结构清晰,肝血窦基本正常,肝细胞索间增生的纤维细胞数量减少,肝细胞有轻微的颗粒变性。结论:青刺果多糖对糖尿病小鼠肝脏具有保护作用。
Objective: To investigate the effects and protective mechanism of Polysaccharide from Prinsepia Utilis Royle on the pathological changes of hepatic tissue induced by alloxan in diabetic mice. Methods : Alloxan was administered intraperitoneally to establish diabetic models in mice.After the modeling, experimental animals were divided into the model group, polysaccharide from Prinsepia Utilis Royle treatment group,positive control group and normal control group. Two weeks and four weeks later, mices were randomly sacrificed,take out of liver, weigh and liver index number were measured,and then fixed into the 4% paraformaldehyde, make slices, the changes of the pathology were observed with Hematoxylin and Eosin staining ( HE ) under light microsc-ope.Results : Polysaccharide from Prinsepia Utilis Royle and positive control group could significantly decrease liver index ( P〈0.05 ) .The pathological and histological observation: liver cell cords of the model mice was inordinate, liver sinusoid broadened and fibrocyte between liver ceils cords was hyperplastic.Fatty degeneration and focal necrosis appeared in liver ceils and there were a large number of inflammatory ceils in necrotic areas and around the central veins. After treated with Polysaccharide from Prinsepia Utilis Royle, structure of liver cell cord became clear, liver sinusoid almost recovered.The number of fibrocyte proliferated reduced and hepatic ceils had granular degeneration slightly.Conclusion : Polysaccharide from Prinsepia Utilis Royle can protect the liver of the diabetic mice.
