肼苯达嗪诱导白血病细胞雌激素受体α(ERα)基因表达的初步研究

摘要 目的研究肼苯达嗪能否诱导白血病细胞ERα重新恢复基因表达。方法应用RT-PCR检测肼苯达嗪处理白血病细胞株HL-60、MOLT-4、Jurkat前后雌激素受体α(ERα)基因表达的变化。结果三种白血病细胞经过肼苯达嗪处理后,ERα恢复mRNA表达。结论肼苯达嗪对白血病细胞的ERα基因具有诱导基因表达的作用,可能使之成为一种新的白血病的去甲基化治疗药物。 Objective To investigate the possibility of reexpression of the estrogen receptor α(ERα) gene in leukemic cell lines (Jurkat, Molt- 4, and HL - 60) treated by hydralazine. Method The variation of expression of ERαmRNA in leukemic cell lines was detected by RT - PCR. Result With the treatment of hydralazine, ERαmRNA was reexpressed. Conclusion Hydralazine can induce reexpression of ERamRNA in leukemic cell lines (Jurkat, Molt - 4, and HL - 60 ), and may be a new demethylation agent.
