Simultaneous Determination of Flavonoid Compounds in Lonicera japonica and its Leaf by High Performance Liquid Chromatography

摘要  目的建立金银花和金银花叶提取物中黄酮类物质槲皮素及木犀草素的含量测定方法。方法采用反相高效液相色谱法,以甲醇-0.02%磷酸(50:50)为流动相,检测波长350nm。结果槲皮素、木犀草素分别在4.0—20,3、8—191μg/ml的浓度范围内与峰面积积分值呈良好的线性关系,它们的加标回收率在95.07%~97.98%范围内,相对标准偏差0.32%~3.34%。结论此方法简便、灵敏、快速,可用于金银花药材中黄酮类物质的含量测定。 Objective To establish the method for the determination of both quercetin and lonicerin in Lonicera japonica and its leaf simultaneously. Methods The components of quercetin and lonicerin in Lonicera japonica and its leaf were determined by the high performance liquid chromatographic at detecfon wavelength of 350 nm with methanol-0. 02% phosphoric acid as mobile phase. Results The calibration curves of quercetin and lonicerin between the peak areas and concentrations were linear within their testing ranges of 4.0 - 20 μg/ml and 3.8 - 19μg/ml respecitively, and the recoveries of two compounds were all between 95. 07% - 97.98%, RSD was 0.32% - 3.34%. Conclusion The results showed that a simple, sensitive, precise method can be used to determine the flavonoid compounds in Lonicera Japonica.
