摘要
目的:观察异基因性NK细胞在小鼠体内抗白血病作用。方法:建立荷EL9611(H-2^d)红白血病细胞的CB6F1^H-2b/d小鼠动物模型,30只红白血病小鼠随机分为5组,每组6只。实验1组:每只小鼠输注NK细胞2×10^5;实验2组:每只小鼠输注NK细胞5×10^5;对照1组:未治疗组;对照2组:阿糖胞苷治疗组;对照3组:同基因NK细胞治疗组。以血象、体重、生存时间和病理组织学变化为观察指标并作组间比较。结果:①生存时间两实验组之间比较差异无显著性(P〉0.05)。对照2组、实验组生存时间高于对照1组(P〈0.01);对照2组高于实验组(P〈0.01);实验组高于对照3组(P〈0.01)。②注入EL9611后第10天,原始细胞占有核细胞的百分比两个实验组比较差异无显著性(P〉0.05);对照2组、实验1、2组低于对照1组(P〈0.01);对照2组低于实验组(P〈0.01);实验组低于对照3组(P〈0.01)。③小鼠发病时脾脏明显肿大,肿大脾脏可达正常5-7倍,HE染色后,镜下可见脾脏正常组织结构破坏,有弥漫白血病细胞侵润。结论:在荷EL9611(H-2^d)红白血病CB6F1^H-2b/d动物模型上证实来源于H-2b的NK细胞能延长荷EL9611(H-2^d)红白血病细胞CB6F1^H-2b/d小鼠的生存时间。
Objective To Study the antileukemic activity of donor-derived NK cell in mice in vivo. Methods CB6F1^H-2b/d mouse model of EL9611 (H-2^d) erythroleukemia was developed by injection of EL9611 (H-2^d) cells in tail vein. 30 mice with erythroleukemia were randomly divided into 5 groups (six mice in each group). Experimental groups (2 groups): 2 × 10^5 and 5 × 10^5 NK cells of C57 BL/6^H-2b were injected into mice after injected EL9611 (H-2^d) cells for 5 d in the first and the second experimental groups. Control groups (3 groups): the first group was the one without treatment; the second one was the Ara-c-treated group which Ara-c were injected into mouse at 50 mg·kg^-1 ×6 d followed by infusion of EL9611 (H-2^d) for 5 d; the third one was syngeneic NK cell treated group, CB6F1^H-2b/d NK cells (1× 10^6 ) were injected into CB6F1^H-2b/d mice after injected EL9611 ( H-2^d) cells for 5 d. The effect was assessed by blood routine test, survival time, body weight, and histopathology in the recipients. Results ①There was no significant difference of life span between two experimental groups (P〉0.05). The survival time of the second control group and experimental groups was much longer than that of the first control group (P〈0.01); the survival time of the second of control group was much longer than that of experimental groups (P〈0.01); the survival time of experimental groups was much longer than that of the third control group (P〈0.01). Conclusion NK cell (H-2^b) can delay erythroleukemia (EL9611, H-2^d) CB6F1^H-2b/d death time in erythroleukemia (EL9611, H-2^d) CB6F1^ H-2b/d model.
