摘要 目的:观察蜈蚣对宫颈癌Caski细胞生长的抑制作用。方法:实验于2006-06/09在三峡大学医学院免疫学重点学科实验室完成。运用体外细胞培养技术,以不同浓度(8,16,32g/L)的乙醚、乙醇蜈蚣提取物作用于培养的Caski细胞48h,采用MTT法测定细胞代谢率,以流式细胞术观察DNA含量和凋亡的变化情况。结果:①细胞形态由梭形逐渐变圆,胞浆皱缩,体积缩小,细胞数量明显少于阴性对照组。②MTT以浓度依赖方式降低吸光度值,与对照组相比抑制率增高显著。③细胞周期各时相DNA含量改变明显,G0~G1期细胞显著降低(由最初64.0%降至最低37.2%),而S,G2~M期细胞明显上升;凋亡率显著增高,阳性对照药物顺铂组也有上述类似改变。结论:中药蜈蚣乙醚、乙醇提取物在体外对宫颈癌Caski细胞的生长有明显地抑制作用,机制与影响癌细胞的DNA合成,阻止瘤细胞的分裂增殖,并促进其凋亡有关。呈现一定的量效和时效相关性。 AIM: To observe the inhibitory effects of centipede extraction on proliferation of cervical cancer Caski cell. METHODS: The experiment was conducted in Lab of Immunology Keystone Subject of Medical College, China Three Gorges University from June to September 2006. By in vitro cell cultural technology, Caski cells received centipede extraction by aether or ethanol at different concentration (8,16,32 g/L) for 48 hours. Cell metabolism rate was measured by MTT. The DNA content and apoptosis were observed by flow cytometry. RESULTS: ①The cell shape changed from fusiform to round, endochylema shdnked, cellular volume contracted, cell population decreased obviously than negative control group. ②The absorbance of MTT degraded with concentration, and compared with control group, the inhibitory rate increased significantly. ③The DNA content changed significantly in each phase of DNA cycle, such as the number of Go-G1 cells decreased significantly (from the first 64.0% to the lowest 37.2%), S,G2-M cells number and the apoptosis rate increased obviously. DDP had the same change, too. CONCLUSION: Centipede extraction by aether or ethanol can markedly inhibit the growth of Caski cell, which is related to inhibitory DNA synthesis, block of division growth of cancer cell and enhancement of apoptosis. The inhibitory effect is depending on quantity and time.
