摘要
目的:应用酵母双杂交技术筛选获得与核因子κB(NF-κB)p50亚基同源结构域(RHD)相互作用的多肽。方法:应用酵母双杂交技术,以NF-κB p50 RHD为诱饵,筛选16肽cDNA文库,寻找与其相互作用的多肽,应用β-gal试验、酵母交配试验及一对一酵母回复性杂交等方法筛除假阳性克隆,确定阳性克隆。结果:经酵母双杂交及反复的排查,共获得8个阳性克隆。通过GenBank进行检索,未发现与之相匹配的蛋白序列。结论:获得8个与NF-κB p50 RHD相互作用的新型多肽,多肽的功能需进一步验证。
AIM: To obtain the polypeptides interacting with NF-κB p50 Rel homology domain (RHD) by yeast two-hybrid technique. METHODS:Using NF-κB p50 RHD as bait, the polypeptides interacting with NF-κB p50 RHD were screened from a 16-pepUde cDNA library by yeast two-hybrid technique. The false positive clones were screened out but the positive clone were identifyied by β-GAL assay, yeast mating, and one to one yeast two-hybrid method. RESULTS: 8 positive clones were obtained. The gene sequences of the eight polypeptides were different from each other and their homologous proteins were not found in GenBank. CONCLUSION: 8 novel polypeptides interacting with NF-κB p50 RHD were obtained, but their functions need to be validated in further experiments.
