摘要
以紫花苜蓿下胚轴、子叶、茎和叶为外植体诱导产生愈伤组织,用微波辅助提取的方法提取其中的皂苷类物质并用比色法测定其皂苷含量。经测定:叶愈伤组织中皂苷含量较多,达到0.41%。以紫花苜蓿叶为材料分别用索氏提取法、微波辅助提取法和超声波提取法提取其中的皂苷类物质,结果表明:微波辅助提取法与其它两种方法相比较,在溶剂的需求量、提取时间和能量的消耗方面优于其它两种方法,它的有效提取时间为2min。
Hypocotyl, cotyledon, stem and leaf of alfalfa were taken as the explants to be induced callus in this paper. Saponin from callus was extracted using microwave assisted extraction (MAE) and quantified using colorimetry.The results show that callus induced from the leaf contains more saponin than others, up to 0.41%.In addition, soxhlet extraction method, MAE and ultrasonic extraction method were respectively used to extract saponin from the leaves of alfalfa.MAE proved to be superior to the other two methods considering the amounts of solvent required, extraction time and energy expended. The efficient extraction time of MAE is 2 min.
