摘要
目的:研究活血化瘀汤在骨折愈合过程中对骨钙素(bone gamma carboxyglutamate protein,BGP)和Ⅰ型胶原(CollagenⅠ)表达的影响,探讨该方在骨折愈合中对软骨内成骨的作用。方法:选用雄性SD大鼠60只,造成左胫骨中段闭合折骨标准的骨折模型,术后采用髓内针固定。随机分成中药组(麻醉苏醒后灌服活血化瘀汤,n=30)和对照组(麻醉苏醒后灌服等量生理盐水,n=30),分别于灌胃后3、5、7、14、21d腹主动脉取血,然后取骨折端上下各0.5cm长的一段骨质。应用Elisa技术,动态观察骨钙素和Ⅰ型胶原表达的变化。结果:血清骨钙素、Ⅰ型胶原在骨折愈合过程中表达持续增高,且表达具有一致性;活血化瘀汤组血清骨钙素、Ⅰ型胶原的表达均较生理盐水组明显(P<0.05)。结论:活血化瘀汤能调控骨钙素和Ⅰ型胶原表达,可能通过影响大鼠骨折愈合过程中软骨内成骨,促进骨折愈合。
Objective:To observe the effects of Huoxue Huayu decoction on the expression of bone gamma carboxyglutamate protein (BGP) and Collagen I in rat fracture healing, nethods:Kirschner' s wires were fixed in the bone marrow of the midshaft tibia with standardized closed fracture in 60 SD rats to establish the fracture models. The rats were randomly and equally divided into Huoxue Huayu decoction group served as Huoxue Huayu decoction and normal saline group served as saline after fracture. The specimens of serum and bony calluses were collected at 3,5,7,14,21 days to detect the level of serum BGP and Collagen Ⅰ after fracture. Results: All 60 rats were involved into the results of analysis. The expression of BGP and Collagen Ⅰ in all serum and the bony callus increased during fracture healing. It was significantly higher in Huoxue Huayu decoction group than that of normal saline group ( P 〈 0. 05 ). Conclusion: Huoxue Huayu decoction may influence the proliferative ability of osteocytes via increasing the level of BGP and Collagen Ⅰ in serum and bony callus of rats with early fracture healing, and thus promote fracture healing.
