摘要
简单节杆菌3-甾酮-1-脱氢酶(KSDH),是甾体母核降解的关键酶,属于黄素蛋白类,并且是一种膜蛋白。膜蛋白的高效表达一直是一个难题。本文尝试利用三种不同的大肠杆菌表达体系,pET-30a/BL21(DE3)、pET-40b(+)/BL21(DE3)和pTrc99A/JM109对C1,2位脱氢酶进行诱导表达,并对蛋白的表达量和酶活力进行比较分析,确定出最适的表达系统。
3-ketosteroid-Δ^1-dehydrogenase (KSDH) belongs to flavoprotein and membrane protein, which is the key enzyme in the degradation of the nucleus of steroids. The overexpression of membrane protein is a difficult problem all the time. In this research, the three different expression systems of E. coli, pET-30a/BL21 (DE3), pET-40b( + )/BL21 (DE3), pTrc99A/JM109 were used to express the KSDH. By comparing the expression level and enzyme activity of target product, the optimum system was finally determined.
