异丙嗪对脑缺血-再灌注损伤大鼠线粒体功能的影响

摘要 目的观察异丙嗪对脑缺血-再灌注损伤大鼠线粒体酶的活性、线粒体内钙含量和线粒体内膜跨膜电位(Δψm)的影响.方法建立大鼠脑缺血-再灌注损伤模型,随机分为对照组、脑缺血组、异丙嗪1组(P1组)和异丙嗪2组(P2组),分别于术后1、6、24 h分离制备脑线粒体悬液,对线粒体纯度和活性进行酶学测定,以及测定线粒体内钙含量和Δψm的变化.结果(1)脑线粒体琥珀酸脱氢酶(SDH)活性在缺血-再灌注后1、6、24 h均低于对照组和P1组、P2组;(2)脑缺血组线粒体内钙含量明显升高,其中6 h时间点线粒体内钙含量升高最为明显(P<0.01);(3)脑缺血组Δψm明显降低,其中6 h时间点Δψm下降最为明显(P<0.01).(4)异丙嗪对脑缺血-再灌注后线粒体钙超载和Δψm的下降和丧失有明显抑制作用,并呈剂量依赖性关系(P<0.05).结论异丙嗪能够抑制脑缺血-再灌注诱导的线粒体通透性转换,从而具有保护线粒体的功能. Objective To observe the effects of promethazine on the activity of mitochondrial enzyme, the concentration of mitochondrial calcium and mitochondrial transmembrane potential (△φm). Methods The rats with brain ischemia - reperfusion were randomly allocated into four groups (n = 18) - normal control group, brain , ischemia group, promethazine 1 group (P1 group) and promethazine 2 group (P2 group). Cerebral mitochondria were isolated 1 h, 6 h and 24 h after operation respectively. Purity and activities of mitochondria were determined by the succinate dehydrogenase (SDH) activity and the concentration of mitochondrial calcium and △φm were measured. Results (1) The SDH activity of the three time points in brain ischemia group significantly decreased compared with those of normal control group and P1, P2 group (P 〈 0. 01 ). (2) The concentration of mitochondrial calcium of the three time points in brain ischemia group were higher than those of normal control group (P〈0. 01), in which the group of 6h after operation was the m time points in brain ischemia group were significantly decreased compared ost remarkable. (3) △φm of the three with those of normal control group (P 〈 0. 01 ) ; in which the group of 6 h after operation was the most remarkable. (4) Promethazine could significantly restrain Ca^2+ overload of mitochondria and the decreasing loss of the △φm induced by brain ischemia - reperfusion, and took on the dose dependent relationship (P 〈0. 05). Conclusion Promethazine could inhibit opening of mitochondria permeability transition pore and protect mitochondrial functions of brain ischemia - reperfusion injury rats.
