摘要
目的用血清药理学的方法研究炙甘草汤含药血清对兔心肌细胞L型Ca2+通道电流(ICa-L)的影响。方法进行单个心室肌细胞的分离和膜片钳全细胞记录,实验分为对照组和5%、10%、20%、40%空白血清组及5%、10%、20%、40%含药血清组,分别以普通细胞外液及加入上述浓度血清的细胞外液进行灌流,检测各组对ICa-L的影响。结果含药血清各组均可抑制ICa-L,5%、10%、20%、40%含药血清分别将ICa-L峰值从(9.4±1.0)电流密度(pA/pF),降至(7.8±1.0)、(6.7±0.7)、(5.6±0.9)、(5.7±1.1)pA/pF。结论炙甘草汤含药血清可抑制ICa-L,且呈浓度依赖性作用增强,这可能就是炙甘草汤抗心律失常作用的机理。
Objective To investigate the effect of serum containing Zhigancao Decoction on L-type calcium current (ICa-L) in isolated single rabbit's ventricular myocyte. Methods Single rabbit' s ventricular myocyte was isolated with enzyme. The whole-cell pAtch clamp recording technique was used to observe the changes of ICa-L. The ventricular myocytes were divided into 6 groups, normal group , normal serum group, 5%, 10%, 20% and 40% serum containing Zhigancao Decoction group. Results The serum containing Zhigancao Decoction decreased the ICa-L density in isolated rabbit's ventricular myocytes;5% , 10% , 20% and 40% serum decreased the lca.L density from(9.4 ±1. 0) pA/pF to(7.8 ± 1.0), (6.7±0.7 ), (5.6 ±0.9) pA/pF and (5.7±1. 1) pA/pF respectively. Conclusion The serum containing Zhigancao Decoction can decrease the ICa-L in isolated rabbit' s ventricular myocytes in a dose dependent manner, which may be its mechanism for treating arrhythmia.
