不同能量等级钬激光照射对膀胱癌细胞株T-24增殖活性与凋亡的影响 被引量:1

摘要 目的观察不同能量等级钬激光照射人膀胱癌细胞株对膀胱肿瘤细胞增殖活性的影响与诱导肿瘤细胞凋亡中的作用。方法人膀胱癌细胞株T-24细胞以不同等级低剂量钬激光直接照射48h后,应用MTT法观察细胞增殖受抑制情况以及光镜下观察细胞形态学的改变;并根据结果选择适宜的钬激光能量值照射T-24细胞后,采用免疫荧光分析钬激光照射肿瘤细胞株后对肿瘤细胞增殖的抑制情况以及对细胞凋亡情况的影响。结果随着钬激光辐射能量的增大,细胞增殖抑制率上升,呈现出一定的剂量依赖关系。根据预试验结果选取钬激光能量值为800mJ辐射细胞株干预后48h采用普通光镜及荧光显微镜观察细胞形态均提示实验组细胞生长密度降低,与对照组细胞在形态学上差异明显,符合细胞凋亡的改变。结论体外细胞试验结果表明,采用钬激光照射人膀胱癌细胞株T-24,能够抑制肿瘤细胞增殖活性并诱发肿瘤细胞凋亡。 Objective To observe the effect of different energy levels' holmium laser radiate Inducing growth inhibition and aoptosis in T-24 bladder carcinomacell.Methods The human bladder carcinoma cell T24 is treated with holmium laser. The MTT assay outcome is referred to decide the holmium energy for the later experiment. The specific holmium energy that affects cell well is given for the further experiment, and the fluorescent microscope research are performed in 48h to detect the cellular apoptosis. Results The obvious growth inhibition of T-24 cell is found,and it proves that the cell livability gets lower with the increase of exposure holmium energy. It suggests some dosage-dependent( P 〈 0.(15).The light microscope observation shows that the T-24 cells treated by the laser holmium wrinkle and shrink and much less in amount. The fluorescent microscope displays clearly that the trial group cells' change according with the change of cellular apoptosis. Conclusion The growth of human bladder carcinoma cell can be inhibited if radiated by low holmium laser energy.Also,holmium laser can induce the cell apoptosis.
