摘要
目的探讨基质金属蛋白酶-2(MMP-2)和组织金属蛋白酶抑制物-2(TIMP-2)在人脑胶质瘤组织中的表达及其与胶质瘤的侵袭发展、分级之间的关系。方法采用RT-PCR方法对20例脑胶质瘤标本及10例正常脑组织进行了MMP-2、TIMP-2表达情况的检测。结果在高、低度恶性组中,MMP-2及TIMP-2的含量较正常对照组均有明显升高并有显著性差异(P<0.05),且高、低两组之间也有显著性差异(P<0.05)。另外,MMP-2/TIMP-2的比值在各组之间也有显著性差异(P<0.05)。结论MMP-2及TIMP-2对胶质瘤恶性程度的评估有重要意义。随着肿瘤细胞恶性程度的增高,MMP-2及TIMP-2的含量也相应增高,但TIMP-2不及MMP-2增高的明显。
Objective To investigate the expressions of the matrix metalloproteinase-2(MMP-2) and the tissue inhibitor of metalloproteinase-2(TIMP-2) in the neurospongioma of human brain and their roles in the pathogenesis and the grade of gliomas. Methods Reverse transcription polymerase chain reaction (RT-PCR) was used to examine the levels of MMP-2 and TIMP-2 in surgical samples from 20 patients with gliomas and 10 norreal controls. Results The expression levels of MMP-2 in grade Ⅰ -Ⅱ and grade Ⅲ-Ⅳwere increased respectively. The expression leuels of TIMP-2 in these two groups increased respectively. The differences between these two indexes in two groups had statistical significance (P 〈 0.01 ). There was negative correlation in the expression of MMP-2 and TIMP-2 ( P 〈 0.01 ). Meanwhile, the ratio of MMP-2 and TIMP-2 was different in each groups significantely. Conclusion The increased expression of MMP-2 and TIMP-2 in human glioma tissue play an important role in evaluating the malignant degree of human glimas.
