斗鸡新城疫病毒DQ株融合蛋白基因序列分析

摘要 从越南斗鸡暴发的病例中分离出1株新城疫病毒(NDV),命名为贵州分离株(DQ)。采用RT—PCR方法扩增分离株(DQ)的整个F基因片段,并将其克隆到pMD-18T载体中。序列分析结果表明,DQ株F基因分子长为1662bp,编码553个氨基酸。该毒株与国内2株分离株(CH2000、TW2000)之间的亲缘关系较近,其核苷酸和氨基酸序列同源性分别为95.7%-96.6%和96.6%-96.8%;但与LaSota、B1及1748E9等常见毒株的核苷酸和氨基酸同源性仅为84.0%-86.5%和87.2%-89.9%。DQ分离株在F蛋白裂解住点的氨基酸顺序为^112R—R—Q—K—R—F^117,与NDV强毒株特征相符,且具有101处的K(赖氨酸)和121处V(缬氨酸)2个特征性氨基酸,与NDV基因Ⅶ型的特性完全相符。利用MegAlign软件绘制NDV的系统发育进化树表明该分离株为基因Ⅶd亚型。 A strain of newcastle disease virus (NDV) was isolated from affected fighting-cock in Guizhou and designated by the name of DQ strain. The entire fragment of F gene of the isolate was amplified by RT - PCR and further cloned into pMD -18T vector. The sequence analysis showed that the nucleotide sequence of the F fragment was 1 662bp,coding for 553 amino acid. The isolate was closer to two Chinese strains( CH2000 ,TW2000) in genetic relationship and nueleotide and amino acid sequence homologies were found to be 95.7% - 96.6% and 96.6% - 96.8% respectively. But the nucleotide and the amino acid sequence homologies to other NDV strains such as LaSota, B1 and F48E9 were only 84.0%-86.5% and 87.2%-89.9% respectively. The deduced amino acid sequence of the cleavage site of F protein Was^112R - R - Q - K - R - F^117 , conforming to the vindent NDV strains. The isolate possessed KWl and V121 , which was similar to the characteristic of genotype Ⅶ. The NDV systemic evolution tree was depicted by MegAlign software and showed that the DQ strain belonged to gene type Ⅶd.
