山羊胚胎皮肤的发育及β1整合素在发育中的表达 被引量:3

摘要 目的:观察山羊胚胎皮肤发育的过程以及在此过程中表皮干细胞的特异性标志β1整合素的表达状况,以定位表皮干细胞。方法:实验于2006-11/2007-05在西北农林科技大学动物科技学院组织胚胎学实验室完成。取8~18周龄山羊胚胎背部皮肤,用组织学方法,观察山羊胚胎皮肤发育的形态学变化;用免疫组织化学SP法,观察β1整合素在山羊胚胎皮肤发育过程中的表达特征。结果:①表皮于8~9周龄时开始发育,10周龄时表皮由三层细胞构成,在11周龄时可见有原始毛囊的出现,有少量张力原纤维的存在,并发现朗格罕斯细胞,12~13周龄时表皮细胞出现角化层、透明层及颗粒层的分化,可见汗腺,14~15周龄毛球上皮细胞索开始分化为外根鞘、内根鞘的原基和毛发的前体,在毛囊一旁出现皮脂腺,张力原纤维增多、变粗,并发现典型的桥粒结构,16~17周龄时表皮层变薄,毛开始形成,并随着发育逐渐长出体表。18周龄皮肤的结构渐趋成熟。②胚胎发育早期,基底层细胞胞浆内表达β1整合素,随着皮肤发育,β1整合素阳性细胞出现在毛囊隆突区和外根鞘。结论:①山羊胚胎表皮的发生优先于真皮的发生,在胚胎时期处于剧烈的分化阶段。②β1整合素的阳性反应定位于基底层,随着皮肤的发育也表达于毛囊隆突区和外根鞘,表皮干细胞的数量和分布的部位与皮肤的发育特点有关系。 AIM: To observe the development of goat embryonic skin and the expression of integrin β1, so as to locate the epidermal stem cell. METHODS: The experiment was carried out in the Laboratory of Histology and Embryology, College of Animal Science and Technology, Northwest Agricultural & Forestry University from November 2006 to May 2007. The back skin of goat embryo at 8-18 weeks old was selected as the samples. Morphological changes of skin development were observed by histological methods. Immunohistochemical staining was carried out to observe the expression state of integrin β1 during skin development. RESULTS ①Epidermis of goat embryo began to develop from the 8^th to 9^th week, and at the 10= week, it was composed of three layers cells; folliculus pili appeared at the 11^th week with few tenofibril and Langerhans cells; from weeks 12 to 13, epidermal cells began to differentiate into cuticular layers, diaphanotheca and granular layers, sweat glands formed at the same time; epidermal cells of hair bulb began to differentiate into the fundament of outer root-sheath and internal root sheath, and the antecedent of hairs, meanwhile, sebaceous gland was formed on one side of the folliculus pill, bridge corpuscle appeared as the tenofibril was thickening; from the 16^th to 17^th weeks, the cuticular layer became thin, and the structures of skin appeared and became mature at the 18^th week. ②Integrin β1 was expressed in the endochylema of basal layer cells during the early period of embryonic development; as the skin developed, positive cells of integrin β1 appeared in the swelling area of folliculus pill, and the outer root-sheath. CONCLUSION: ①The development of goat embryonic epidermis, which differentiates exquisitely during the embryo phase, is priority to that of corium. ②Positive reactions of integrin β1 is located in the basal layer. As skin growing, it is also expressed in the swelling areas of folliculus pili and the outer root-sheath. The amount and distribution of epidermal stem cells are associated with the development traits of skin.
