骨髓间充质干细胞载体复合膜修复皮肤缺损的实验研究 被引量:1

摘要 目的探讨应用自体骨髓间充质干细胞的羊膜载体复合膜(羊膜-明胶-壳聚糖膜)植入皮肤全层缺损处对真皮的再生和重建速度的影响。方法抽取16只新西兰大耳白兔的骨髓间充质干细胞,经体外分离、培养并于其背部脊柱两侧制作两个全层皮肤缺损创面、随机分为两组。治疗组为骨髓间充质干细胞和载体复合膜,对照组单纯为载体复合膜。7、14、21天取材观察结果。采用大体观察、HE染色、Masson染色、I型胶原免疫组织化学染色、电镜观察、墨汁灌注法等化学染色动态观察创面愈合情况。结果治疗组在7、14、21天时新生真皮明显较对照组相应时间点的厚、活跃的I型胶原阳性细胞数多、电镜下粗面内质网扩张的成纤维细胞、血管和胶原纤维均较对照组的多。结论用含骨髓间充质干细胞的载体复合膜修复全层皮肤缺损,能加速真皮的修复和重建。 Objective Use amnion carrier compound which contain self marrow stem cell(MSC)plant on membrane(amniotic membrane gelatin-chitosan) the deep-partial thickness wounds, For discuss if this method can deep influence renascence and regenerate of the dermis, Methods MSCs were isolate from 16 rabbit's marrow. The MSCs were culture and purified in vitro.There are two wounds on the beck of the each experimental animal. Experimental groups are consisted of MSCs and amniotic membrane-gelatin-chitosan,Control groups are consisted of amniotic membrane gelatin chitosan. Histological examination was performed to evaluate the velocity and the quality of the wound. Immunohistochemical examinations were carried out to detect the positive staining Ⅰ type collagen. The dennis tissue was observed by electron microscopic. HE dyeing after put ink perfusion into blood vessel. Results The depth of the renascence dermis, the number of the active type Ⅰ collagen masculine cell, the number of the observe from the electron microscopic outstretched endoplasm' s fibroblast, the number of the blood vessel, all these Experimental group are better than control group. Conclusion Use amnion carrier compound membrane which contain self marrow stem cell(MSC) repaired the skin wounds. They can influence the rapidity renascence and regenerate of the dermis.
