土壤中产碱性蛋白酶细菌的筛选及产酶条件优化 被引量:6

摘要 为筛选碱性蛋白酶的高产菌株,利用酪蛋白水解圈作为筛选模型,从沈阳化工学院附近农田土壤中筛选得到了产碱性蛋白酶能力较高的细菌B1.对其产酶条件进行优化,最终得到培养基中最佳碳源为质量浓度60 g/L葡萄糖,最佳氮源为质量浓度40 g/L NaNO3,最适初始pH值为9.0,最适溶氧量为50 mL摇瓶装20 mL培养基,最适接种量为质量浓度200 g/L,且培养基中添加质量浓度为0.5 g/L的K+有利于菌株B1产碱性蛋白酶.该方法简便、直观,很适合大量、快速筛选. In order to screen a bacterium which can produce alkaline protease, Casein hydrolyzation circle was used as screening model in this paper. The bacterium B1 was achieved by screening from the soil near the Shenyang Institute of Chemical Technology. After the optimization of the condition, the capability of B1 was improved, the optimal condition is: carbon source is glucose 6 %; nitrogen source is NaNO3 4 % ,the pH is 9.0; the appropriate load is 20 mL culture medium in 50 mL flask; quantity of inoculation is 200 g/L. when K^+ is 0.5 g/L in the culture medium, the capacity of B1 can be improved. This method used in this paper is very simple and quick.
