摘要
目的:研究肺癌细胞生存素(survivin)基因表达及其与放疗敏感性间的关系,探讨survivin作为预测肺癌放疗敏感性分子标志物的可能性。方法:设计合成survivin基因引物,并对肺癌组织标本行RT-PCR以检测其survivin表达。对原代培养肺癌细胞行2.0Gy X线照射后,采用Cell Proliferation Kit I(MTT)法进行细胞活力检测。结果:survivin在肺癌组织中阳性表达率较高,为60.9%,而在正常肺组织中阳性表达率仅为14.3%。肺癌细胞经2.0Gy的X线照射后,细胞增殖明显受抑,存活率下降。但survivin阳性肺癌细胞的存活率(40.0%~70.0%)显著高于survivin阴性肺癌细胞(20.0%~50.0%)。结论:肺癌高表达survivin,并与其放射敏感性有关。X线照射对癌细胞生长有明显抑制作用,但survivin阳性的癌细胞对X线照射有一定耐受性。检测肺癌细胞survivin表达有望成为预测肺癌对放疗不敏感的辅助指标之一。
Objective To investigate the survivin gene expression and its relationship with radiosensitivity in lung cancer cells,and to analyze the possibility of survivin as a molecular marker for prediction of lung cancer radiosensitivity.Methods The specific primers of survivin gene were designed and the survivin expression in the specimens of lung cancer was identified by RT-PCR.The lung cancer cells were radiated by 2.0 Gy of X-ray,then the cell viability was calculated by MTT assay.Results The over-expression of survivin was showed in lung cancer with the positive rate of 60.9%,whereas 14.3% in normal lung tissues.After radiated by 2.0 Gy of X-ray,the growth of lung cancer cells was inhibited with the decrease of cell viability below 60.0%.But the cell viability of survivin-positive group(40.0%-70.0%) was obviously higher than that of survivin-negative group(20.0%-50.0%) .Conclusions The lung cancer over-expressed survivin,which related to the cell radiosensitivity.The radiation of X-ray could inhibite the growth of cancer cells,but there was a tolerance to radiation in the cancer cells with positive survivin expression.The survivin expression may be a useful marker to predict the radiosensitivity of lung cancer.
