摘要 近年来的研究发现,单核细胞趋化蛋白1(MCP-1)是一种极为强烈的单核细胞趋化因子。本研究着重观察了极低密度脂蛋白(VLDL)和氧化修饰的极低密度脂蛋白(OX—VLDL)对单核细胞和巨噬细胞MCP-1 mRNA表达的影响。在单核细胞和巨噬细胞的培养基中分别加入25μg/ml的VLDL或OX—VLDL,培养24h后提取单核细胞和巨噬细胞总RNA,用MCP-1寡核甘酸探针,进行Northern blot分析。结果显示单核细胞和巨噬细胞都能表达MCP-1 mRNA,OX-VLDL能增强单核/巨噬细胞MCP-1 mRNA的表达。这提示单核和巨噬细胞能促进单核细胞迁入内皮下间隙,而OX-VLDL能增强这种作用。 The effect of oxidatively modified very low density lipoprotein (OX-VLDL) on the MCP-1 mRNA expression by monocytes and macrophages was sludied. The total RNA was extracted flom monocytes and macrophages, which were exposed to OX-VLDL, and VLDL for 24 hours respectively, using guanidmium isothiocyanate method. MCP-1 mRNA expression in monocytes and macrophages was examined by Northern blot analysis using a 7 32p-end-labeled 35-mer oligonucleotide probe. The results revealed that the MCP-1 mRNA expression was markedly increased by a 24-h exposure to OX-VLDL, whereas the MCP-1 mRNA expression was slightly increased, when exposed to VLDL. It was concluded that either monocytes or macrophages can express MCP-1 mRNA, and OX-VLDL induce stronger MCP-1 mRNA expression. It suggests that monocytes or macrophages induce migration of monocyte into subendothelial space by expressing MCP-1. and OX-VLDL enhance the action.
