摘要
发酵条件优化可提高少根根霉菌株8B所产纤溶酶的活性。使用单因素试验和正交试验确定该菌发酵产酶的最佳条件。试验确定最优化发酵条件,培养基:麸皮水5 g/L,尿素5 g/L,胰蛋白胨0.5 g/L,K2HPO4.3H2O0.3 g/L,MgSO4.7H2O 0.15 g/L,pH 5.5,摇床转速160 r/m in,30℃发酵56 h。在此优化条件下培养,8B产纤溶酶活力达到345.41 U/mL,是初始培养基发酵产酶活力的7.52倍。
To improve the activity of fibrinolytic enzyme of Rhizopus arrhizu 8B, effects of single-factor and orthogonal design experiments were investigated. Results showed that the optimium fermentation conditions were as follows: the medum including 5 g/L bran, 5 g/L urea, 0. 5 g/L tryptone, 0. 3 g/L K2 HPO4·3H2O,0. 15 g/L MgSO4·7H2O;pH 5.5,shaker 160 r/min,at 30 ℃, for 56 h. Under the conditions, the enzyme activity reached 345.41 U/mL, which 7.52 times higher than that obtained under the initial conditions.
