摘要
目的:观察粉防己碱对体外培养的人乳腺癌细胞MCF-7细胞株增殖的影响。方法:MTT法检测MCF-7细胞对粉防己碱的敏感性;流式细胞仪检测粉防己碱对MCF-7细胞的细胞周期、凋亡率的影响。结果:随着粉防己碱浓度的增加,作用时间延长,MTT试验中试验组细胞光吸收值降低;流式细胞试验中S期细胞数减少,凋亡细胞比例增加。结论:粉防己碱对MCF-7细胞株的生长有明显的抑制作用,并且这种抑制效应存在剂量和时间的依赖关系;粉防己碱可阻滞MCF-7细胞周期活动,诱导细胞凋亡,且其调亡率和浓度及作用时间呈正相关。
Objective: to observe the effect of tetrandrine on cell proliferation of human breast cancer MCF-7 cell strain in vitro culture. Method: MTT was used to detect MCF- 7 cells sensitivity to tetrandrine and flow cytometry was used to detect the effect of tetrandrine on cell cycle and apoptosis rate. Result: when the density of tetrandrine is increased and lengthened action time, the cell light absorption value is lower in experimental groups in MTT test and cell population also is lower in S stage in flow cytometry test and apoptosis ratio is increase. Conclusion: tetrandrine have the depressant effect to growth of MCF- 7 cell strain and this depressant effect has the symbiosis between dosage and time. Tetrandrine have the function to block the cell cycle of MCF- 7 and induce apoptosis and the apoptosis rate is positive correlation between density and action time,
